RODeo: Everything You Need to Know

From worms to flies, the compounds being tested on pump.science have shown promising results. Now we enter the next phase: mouse trials.

Welcome to the RODeo.

Why RODeo?

Our mice will undergo endurance testing on a rotating rod — the rotaROD — the industry standard for measuring motor function and vitality in aging research. Hence, RODeo.

Each week, mice receiving interventions or placebo, will face off to see which compound enhances performance more.

The Experiment

Groups of mice:

  • Control group: 3 mice

  • Compound group(s): 3 mice per compound tested

How It Works:

Each week, mice face a series of endurance trials on the rotarod — a rotating cylinder designed to test their athletic performance.

The challenge: stay on the rotating rod as long as possible. The rod begins rotating slowly, and speeds up over time. Staying on the rod will test their athletic endurance.

The Study Protocol:

Before official trials begin, mice undergo pre-training at a steady 4rpm to get comfortable with the rotarod.

Weekly Testing Structure:

  • Mice are placed on the rotarod in specified lanes.

  • Each mouse completes 3 trials in one day.

  • Maximum of 15-minute recovery between trials.

  • Speed increases gradually until all mice have fallen off.

  • Time, until they fall off (latency), is recorded.

    • This data can be analyzed to assess motor coordination and balance.

Trial Details:

  • Maximum trial length: 5 minutes

  • Testing continues for 8 weeks

Data collection:

Time on the rod (latency) measures motor coordination and balance. For reliability:

  • All trials happen on the same day.

  • Results averaged across three attempts per mouse.

  • Performance tracked weekly.

  • Data verified within 24 hours

  1. What is the rotarod? A rotating cylinder used to measure motor coordination and balance in research.

  2. Can I watch without participating? Absolutely! Live streams are open to everyone. How are winners determined in prediction markets? Results are based on official data verified and posted on pump.science.

  3. How is the data from the experiments verified? All data is reviewed and verified by our research team, Tracked Bio, within 24 hours of the trials, ensuring accuracy and reliability before results are posted.

  4. Will I have access to historical data from previous RODeo events? Yes, summaries and data from past RODeo events will be available for reference and analysis.

  5. What happens if technical issues affect the live stream or trials? In the event of technical difficulties, all efforts will be made to resume as quickly as possible. Any significant disruptions will be communicated via our official channels. All the experiments will be recorded. If we cannot resume the stream, the full recordings of the experiments will be made available.

  6. How are mouse names chosen? Mouse names are selected through a community voting process, details of which will be shared before the trials begin.

  7. What happens if a mouse doesn’t complete all three trials? Since the main endpoint in rotarod is latency, latency will be recorded as 0 seconds for each trial where the mouse cannot walk at all on the beam when the session starts.

  8. What is the Mouse Tracker data? How is this different than the data from the rotarod? Tracked Bio's TrackedMouseWatcher(TM) data is in-cage of analysis of the mice in their "home" environment. Therefore, we can observe the activity of the mice in an uninterrupted fashion, which overall is a more unbiased data capture. Data that will be generated for this experiment will be distance and speed of the mice. Other features may potentially be included as well.

  9. How do these endpoints correlate to longevity outcomes in the mice? Rotarod is a well-recognized and validated endpoint that assesses motor coordination, balance, and overall neuromotor performance in mice, critical health indicators and functional ability as organisms age.   10. How can you ensure the right data is correlated to the right mouse? Each mouse will maintain the same lane in the rotarod, so identity will be preserved.

  11. How is the experiment designed to ensure the mice are not being harmed? Is the experiment reviewed and approved by an ethics committee? This study was approved by the IACUC. The Institutional Animal Care and Use Committee (IACUC) is a federally mandated committee in the United States responsible for overseeing and approving research involving animals, including mice, to ensure humane and ethical treatment in compliance with federal regulations.
